Ten serial dilutions of the serum in 0.15 M phosphate buffer saline (PBS) pH 7.2 were made and 50ul of these were then titrated with 25ul of 2.5 x 107 SRBC /ml. the test tubes were then incubated at room temperature for 2 h and examined visually for agglutination.
